Precise Oxygen Measurement

The OxyDye is an enhancement available for all OLIS spectrophotometers. Hardware is an LED for flashing at a nanomolar volume of biologically inert dye added to your sample, and the photon counting detector for capturing the absolute photon counts.
  • Capture oxygen levels during absorbance, fluorescence, or CD experiment, or free-standing in a beaker or other vessel in a dark chamber.
  • Software runs entirely independent of any other (spectral) data acquisition program.
  • Pulsed LED under software control excites the sample.
  • A high sensitivity PMT captures the luminescence.
  • Light is transmitted in both directions using light guides.
  • The oxygen concentration is calculated from the luminescence decay (i.e., its lifetime) after the decay profile is fitted to a one exponential and the resulting decay constant (Tau) is converted to O2 concentration using the Stern-Volmer equation.
